چکیده

Leaf rust of barley, caused by Puccinia hordei Otth, is an important foliar disease in most temperate regions of the world. Sixteen major leaf rust resistance (Rph) genes have been described from barley, but only a few have been mapped. The leaf rust resistance gene Rph7 was ®rst described from the cultivar `Cebada Capa' and has proven e€ective in Europe. Previously mapped restriction fragment length polymorphism (RFLP) markers have been used to determine the precise location of this gene in the barley genome. From the genetic analysis of a `Bowman'/`Cebada Capa' cross, Rph7 was mapped to the end of chromosome 3HS, 1.3 recombination units distal to the RFLP marker cMWG691. A codominant cleaved ampli®ed polymorphic site (CAPS) marker was developed by exploiting allele-speci®c sequence information of the cMWG691 site and adjacent fragments of genomic DNA. Based on the large amount of polymorphism present in this region, the CAPS marker may be useful for the marker-assisted selection of Rph7 in most diverse genetic backgrounds.

Molecular mapping of the leaf rust resistance gene rph6 in barley and its linkage relationships with rph5 and rph7.

ABSTRACT The barley cv. Bolivia carries two leaf rust (Puccinia hordei) resistance genes, Rph2 and Rph6, and is the only known source of the latter gene. A resistant line (Bolivia-Rph6) carrying Rph6 only was obtained in the F(4) generation of a cross between cv. Bolivia and the susceptible cv. Bowman via progeny testing with differential isolates of the leaf rust pathogen. Genetic analyses and...

can be easily identified and transferred into appropriate germplasm (Parlevliet, 1976). To date, 16 major raceLeaf rust caused by Puccinia hordei G. Otth is an important disease specific genes (designated as Rph1 to Rph16) for leaf of barley (Hordeum vulgare L.) in many regions of the world. Yield losses up to 32% have been reported in susceptible cultivars. The rust resistance have been identi...

Identification of genetic resources of field resistance to barley leaf rust in local germplasm of cultivated barley

The barley leaf rust has been important in recent years in Iran. In order to identify the genetic resources of resistance to this disease, 207 Iranian barley landraces were studied. The germplasms were investigated at the field of Iraqi-Mahalleh research station in Gorgan as the disease hotspot under natural incidence over three years. The results showed that four genotypes including KC18638 an...

Large intraspecific haplotype variability at the Rph7 locus results from rapid and recent divergence in the barley genome.

To study genome evolution and diversity in barley (Hordeum vulgare), we have sequenced and compared more than 300 kb of sequence spanning the Rph7 leaf rust disease resistance gene in two barley cultivars. Colinearity was restricted to five genic and two intergenic regions representing <35% of the two sequences. In each interval separating the seven conserved regions, the number and type of rep...

A large rearrangement involving genes and low-copy DNA interrupts the microcollinearity between rice and barley at the Rph7 locus.

Grass genomes differ greatly in chromosome number, ploidy level, and size. Despite these differences, very good conservation of the marker order (collinearity) was found at the genetic map level between the different grass genomes. Collinearity is particularly good between rice chromosome 1 and the group 3 chromosomes in the Triticeae. We have used this collinearity to saturate the leaf rust re...
